It is within walking distance from the S.Lucia Train Station. However you do have to cross two bridges which could be hassle if you are carrying heavy bags. I walked it with my wife carrying one heavy bag each and a backpack. I'd do the same again.
We walked the next morning over to the Maritima (spelling??) cruise ship station.It is a longer walk...not sure if there was a shorter route but if I were to do it again I'd walk to the taxi (car) station (short walk form the hotel) and grab one to the ships...
Although the overall service and room was good, welcoming has been scandalous. The person at reception desk has been unable to get the authorization for my credit card (visa gold), even after a telephone check with my bank in France (I had to call the bank with my own mobile phone. additional cost for me = 10 ?); I have been obliged to withdraw cash (with the same card) from a proximate ATM to pay the hotel. It has severely compromized my first day to Venezia : All other payements with the same card during my stay were OK, so I deeply suspect the card terminal of the hotel had a problem. I received absolutely no help from the girl at the desk, and she was absolutely suspicious. Making a reservation at a Sofitel hotel and being a frequent business traveller, I was expecting for more care to customers. I will never go there again.

The location is great - just a throwing stone from Piazzale di Roma and walking distance from Train Station.
The room is nice....
The problem is the attitude of the staff !
The frontliners were bit arrogant, no smile and just like a robot.
